Open iTunes (Windows) or Finder (Mac), go to Device Preferences/Settings, and use the "Delete Backup" button. This ensures the database is cleaned up properly.
If your system drive (C: drive or Mac SSD) is full, you might want to move the MobileSync backup to an external drive. You cannot simply drag the folder; you must create a symbolic link (symlink) so the computer knows where to look.
